Bruton's tyrosine kinase (Btk) was originally identified as a tyrosine kinase that autophosphorylates in response to activation of the B cell receptor, but it has also been implicated in signalling by several receptors, including the B cell receptor, platelet-activating factor, and the Fc-epsilon-R1. To define its physiological role in signal transduction, we studied receptor-induced cellular signalling in fibroblasts derived from Btk-deficient (Btk(-/-)) mice and the ability of these cells to migrate in response to different growth factors. We show that Btk(-/-) fibroblasts proliferated normally in response to growth factors and in response to adhesion to fibronectin. Although the Btk(-/-) fibroblasts spread on fibronectin, adhesion to this matrix was impaired compared with that of wild-type (WT) fibroblasts. WT and Btk(-/-) fibroblasts responded similarly to platelet-activating factor (PAF) in terms of cellular signalling and cellular migration. We show that Btk(-/-) fibroblasts also migrated in response to fibroblast growth factor, epidermal growth factor, and hepatocyte growth factor, despite a low level of phosphorylation of the p85 regulatory subunit of phosphatidylinositol 3-kinase and impairment of cell migration in response to these growth factors. These results define the primary role of Btk in the B cell receptor-dependent signalling pathway and demonstrate that Btk is not required for the fibroblast growth factor, epidermal growth factor, or platelet-activating factor signalling pathways.Q:
